Job Description

Telomeres represent potent signaling detectors for DNA damage in the cell, residing at the ends of our chromosomes. Containing only 1/3000th of the total DNA in the cell, they monitor the number of cell divisions and protect us from uncontrolled cell division that can lead to cancer. Telomeres consist of a complex assembly of repetitive DNA, cellular histone and DNA repair factors, and RNA transcribed from the telomeres themselves termed TERRA. Following transcription of TERRA, some of this RNA remains annealed to the telomere in the form of R-loops. Unresolved R-loops represent one form of DNA damage, yet they may play a central role in forming t-loops, a looped arrangement of the telomere in which the very DNA end is folded back to anneal to internal telomeric sites. The goal of these studies is to use a combination of electron microscopy, light microscopy, cell biology, and biochemical methods to investigate the location, frequency and role of R-loops at the telomere. The relevance of R-loops to human disease will be a central focus. This will be done using mammalian tissue culture and cells exhibiting the ALT phenotype. The person in this position will carry out these experiments, be engaged in collaborations with colleagues in the US and abroad and be exposed to new technologies including cryoEM.

Environment:

The Lineberger Comprehensive Cancer Center (LCCC) is one of the most vibrant and active Cancer Centers in the US and is nestled among the departments of Biology, Biochemistry, Cell Biology, Immunology, Microbiology and Pathology. LCCC is a center for many core facilities which provide access to state-of-the-art technologies from light and electron microscopic imaging, mouse genetics, advanced sequencing, cryoEM, and many biophysical technologies. LCCC has one of the largest NIH funded postdoctoral training grants and due to this we have a very active program for fostering, mentoring and aiding postdoctoral fellows in their career advancement. This includes postdoctoral retreats, postdoctoral seminars, seminars on career development and the spectrum of possibilities for further scientific opportunities. The Griffith laboratory has a stellar history of collaborations, both with colleagues at UNC and nationally/internationally. We have collaborated with 90 different NIH-funded laboratories and generated more than 120 peer reviewed publications with other groups. Thus our postdoctoral fellows are provided with many opportunities to expand their training through engaging in one or more outside collaborations, an effort which is also designed to provide additional publications for their resumes.

Qualifications: The ideal candidate should have a PhD in biochemistry or cell biology or a related field and expertise in at least one of the following areas:

Biochemistry: DNA and protein purification, analysis of unusual DNA structures,

DNA/RNA processing and extraction; protein engineering, protein expression in insect

systems.

Virology: experience in Herpesvirus growth, genetics, and gene expression.

Cell Biology: tissue culture growth of human and murine cells, treatment of cells in culture with

siRNA, oxidizing agents, and over expression of plasmids following transfection.

Biological imaging: experience in imaging cells in culture with light microscopic methods, fluorescent antibody staining, experience in imaging macromolecules or cells by electron microscopy.
